A gifted teenager named Peter Parker is given spider-like powers when he is bitten by a radioactive spider during a school field trip to Oscorp. After the death of his Uncle Ben at the hands of a burglar, Parker comes to his own terms of becoming a masked superhero named Spider-Man, learning to balance his crime-fighting career with his new enrollment at Horizon High, a school for students with genius-level intellect.

- The promotional artwork and toyline created for season one had character designs that were slightly different from the show.
- This is the case for all of the villains who look nothing like their promotional artwork.
- A huge criticism of the show, mainly season one, was how it used science as an explanation to solve a majority of the problems that occurred. This was toned down later on.
- Despite being announced as a reboot of Ultimate Spider-Man, it is not inspired by the Ultimate Spider-Man comic book series like the previous show..
- The Spider-Island story arc was originally planned for Ultimate Spider-Man before the production staff knew it's fourth season would be it's final one.
- The Rhino in both shows is a mutant instead of a man in a suit or armor.
- Season 1 takes place after the first two seasons of the Guardians of the Galaxy Animated Series. The episodes of Guardians of the Galaxy: Mission Breakout! featuring Spider-Man (Back in The New York Groove and Drive My Carnage) take place sometime after the episode Venom. Spider-Man already knows Iron Man by that point, referencing Stark Expo, and Max has the V-252 samples that bonded to Flash and the football players in the Horizon High Think Tank.
- When seeing Thanos in Back in The New York Groove, Spider-Man mentions seeing him attack New York before which happened in the season 1 finale of Guardians of the Galaxy.
