New York City (or just New York) is the main setting of Marvel's Spider-Man. New York City is made up of five boroughs (Manhattan, Brooklyn, Queens, Staten Island, and the Bronx). Judging by the locations seen and mentioned in the series only Manhattan and Queens have been featured in the show.

- The New York City in this series appears to be more technologically advanced out of other Spider-Man and Marvel Comics animated series.
- Some locations are altered from their appearance in the real-life New York City. For example, Spider-Man, Spy-D, and Ghost-Spider should not have been able to reach Liberty Island by gliding in Spider-Island Part One.
- The shot of Manhattan from Horizon High Part One appears to be the same model used in the previous series Ulitmate Spider-Man, but modified to match the art style of Marvel's Spider-Man.
